Phosphatidylinositol 3-kinase phosphorylates the inositol ring of phosphatidylinositol at the 3-prime position The enzyme comprises a 110 kD catalytic subunit and a regulatory subunit of either 85 55 or 50 kD This gene encodes the 85 kD regulatory subunit Phosphatidylinositol 3-kinase plays an important role in the metabolic actions of insulin and a mutation in this gene has been associated with insulin resistance Alternative splicing of this gene results in four transcript variants encoding different isoforms [provided by RefSeq Jun 2011]

The protein encoded by this gene is part of the nuclear lamina a two-dimensional matrix of proteins located next to the inner nuclear membrane The lamin family of proteins make up the matrix and are highly conserved in evolution During mitosis the lamina matrix is reversibly disassembled as the lamin proteins are phosphorylated Lamin proteins are thought to be involved in nuclear stability chromatin structure and gene expression Vertebrate lamins consist of two types A and B Alternative splicing results in multiple transcript variants Mutations in this gene lead to several diseases Emery-Dreifuss muscular dystrophy familial partial lipodystrophy limb girdle muscular dystrophy dilated cardiomyopathy Charcot-Marie-Tooth disease and Hutchinson-Gilford progeria syndrome

The protein encoded by this gene is a component of the eukaryotic translation initiation factor 4F complex, which recognizes the 7-methylguanosine cap structure at the 5 end of messenger RNAs. The encoded protein aids in translation initiation by recruiting ribosomes to the 5-cap structure. Association of this protein with the 4F complex is the rate-limiting step in translation initiation. This gene acts as a proto-oncogene, and its expression and activation is associated with transformation and tumorigenesis. Several pseudogenes of this gene are found on other chromosomes. Alternative splicing results in multiple transcript variants.

The protein encoded by this gene belongs to the family of X-pro-aminopeptidases that utilize a metal cofactor and remove the N-terminal amino acid from peptides with a proline residue in the penultimate position This protein has been shown to localize to the mitochondria of renal cells and have a role in ciliary function Mutations in this gene are associated with nephronophthisis-like nephropathy-1 Alternatively spliced transcript variants encoding different isoforms have been noted for this gene however expression of some of these isoforms in vivo is not known
